The CC4CARB imidazole library (CARB011) had notable antimicrobial activity. Do you believe it warrants additional follow-up analogs? RTI-CARB11-099 showed interesting broad antimicrobial activity. View the CARB011 series here cc4carb-collection.org
A highlight of CC4CARB’s vancomycin–amine library is RTI-CARB8-027, which showed antimicrobial activity across multiple strains including E. coli BW25113 and E. coli efflux-deficient mutant (tolC). This analog, and others, exhibited extraordinary MICs on S. aureus (0.125 µg/mL).
Four positions on the vancomycin scaffold were explored using amine conjugates to penetrate the GN cell wall. Several active candidates emerged against GN bacteria. To view the compound structures/MICs for the CARB008 series, go to cc4carb-collection.org
A key highlight of CC4CARB’s spiroindoline library is the compound, RTI-CARB6-106, which showed broad-spectrum antimicrobial activity across several Gram-negative organisms as well as S. aureus. To view the compound structures/MICs, go to cc4carb-collection.org
CC4CARB synthesized 257 compounds in the spiroindoline library (CARB006). Several compounds produced in CARB006 showed broad spectrum activity with some having a narrow spectrum.

CC4CARB synthesized 103 compounds in the sulfonylpyridine library (CARB004). Several compounds produced in CARB004 showed antimicrobial activity against the E. coli efflux (tolC) JW5503-1 strain.

Over 20% of the vancomycin–siderophore conjugates in the CARB001 series achieved MIC values of 1–2 µg/mL against A. baumannii JMI #1186154 (ACB strain) that could be proposed for further preclinical studies. View the structures and associated MICs here cc4carb-collection.org
A highlight of CC4CARB’s vancomycin–siderophore library is RTI-CARB1-007, that showed antimicrobial activity across multiple strains including E. coli BW25113, an E. coli efflux-deficient mutant (tolC), P. aeruginosa PAO1, & an efflux pump–deficient P. aeruginosa strain (PAM1626)
Four positions on the vancomycin scaffold were explored using simple alkyl chains to PEG to click-chemistry spacers to penetrate the GN cell wall. Many active candidates emerged against GN bacteria. To view the compound structures/MICs, go to cc4carb-collection.org
CC4CARB has published the scaffold-libraries relating to vancomycin analogs (siderophores and amines), pyrimidinodiazepinoes, pyridinodiazepiones, sulfonylpyridines, spiroindolines, imidazoles, and a metal-organic framework. View the collection here:

As 2024 comes to a close, CC4CARB looks forward to serving the global antimicrobial community for another year. CC4CARB will be releasing compound plates in 2025. Watch for announcements from us with all the details on how to request these compounds.
CC4CARB will be releasing our first plate of compounds soon. Watch for our upcoming announcement with all the details for requesting these compounds.
If you have a novel scaffold idea or wish to donate compounds to the collection, please reach out to us at [email protected].
CC4CARB is looking forward to attending conferences in 2024/2025 to showcase the program’s progress and connect with additional organizations in antibiotic discovery. We'd love to schedule a face-to-face meeting with you. Let us know which future conferences you plan to attend.
With nearly 50 scaffolds accepted, CC4CARB is now interested in expanding our collection into other areas such as natural products, conjugates, potentiators, and AI based technology. If you have a proposal idea to discuss with the CC4CARB PI, reach out to us at [email protected].
