What has INB-619 shown so far in vitro? ✓ Deep B cell depletion in lupus donor samples ✓ Selective gamma-delta T cell activation and expansion ✓ Minimal CRS-associated cytokines All preclinical, in vitro results. Next question: Does the biology translate in vivo? $INAB #TCellEngager #Autoimmune

Two lupus donors. Two different immune responses. Same result. INB-619 expanded Vδ2 T cells in one donor and Vδ1 in the other. Both achieved complete B cell depletion by day 4. That’s the rationale for pan-gamma-delta engagement. Preclinical, in vitro data. $INAB #Lupus #TCellEngager

Not all gamma-delta T cells work the same way. Vδ1 cells primarily reside in tissues; Vδ2 cells circulate in the blood and lymphatic system. INB-619 is designed to activate and expand both, plus other subsets, to target B cells across all compartments. Kate explains how. Preclinical data. $INAB #TCellEngager

Gamma-delta T cells are powerful but scarce. INB-619 is designed to overcome that challenge with a built-in domain that both activates and expands these cells. Preclinical studies showed consistent potency despite varying starting cell levels. $INAB #GammaDeltaTcells #TCellEngager

CAR-T demonstrated the potential of deep B cell depletion to reset the immune system, but requires complex manufacturing and chemotherapy. INB-619 aims for similar activity with an off-the-shelf T cell engager. No personalized cell manufacturing or lymphodepletion. $INAB #TCellEngager #Autoimmune

Most T cell engagers activate T cells through CD3, which can trigger broad immune activation and potentially serious side effects. INB-619 takes a different approach: selectively engaging gamma-delta T cells. Same disease target. Different T cell. $INAB #GammaDeltaTcells #TCellEngager

Three engagers, one target class. Two work through CD3; INB-619 works through the gamma-delta receptor.

Cancer and autoimmune disease have very different risk-benefit equations. Most T cell engagers broadly activate the immune system, more like a sledgehammer. INB-619 is designed to be more like a scalpel, selectively engaging gamma-delta T cells for a potentially more precise approach. $INAB #Autoimmune #TCellEngager #Immunology

Why the standard CD3 approach creates a narrow therapeutic window.

In 2021, Georg Schett at the University of Erlangen-Nuremberg published a landmark study, something the field had not seen before. He used CD19 CAR-T to deplete B cells in lupus patients. They went into drug-free remission. The idea: wipe out the misfiring B cells and the immune system restarts from a healthier place. It has since been repeated across several autoimmune diseases. Why CD19? Because it is the broadest target found across B cells at every stage of their life, from early precursors to mature memory cells. Other targets miss earlier stages. Dr. Schett has never seen another target after CD19. That is the target INB-619 is built around. #Autoimmune #CD19 #Immunology $INAB

More than 24 million Americans have an autoimmune disease. In some of them, the cause traces to one cell type: B cells that have turned on the body. Lupus: 200-350k Systemic sclerosis: ~100k Myasthenia gravis: 80-100k Inflammatory myopathy: ~75k Deplete those B cells and the immune system can reset. That's the thesis we're building on. $INAB

~1 - 5% of your white blood cells are gamma-delta T cells. Most of immunology looked past them for decades. They kill on sight. They remember. And they read stress signals on a cell's surface rather than needing to be trained on a target. That combination is rare. $INAB

Pharma committed $8.3B to autoimmune T cell engagers in 2026. Five deals. Nine months. Two were preclinical at signing. The modality is being pursued before the clinical data exists. The open question: which T cell should you engage? $INAB

This month we welcomed Oxana Polyakova, PhD, as SVP of Research & Development. She has spent 20 years building T cell engagers. So what exactly is one? A T cell engager is an off-the-shelf protein with two ends. One end recognizes a T cell. The other recognizes the target. It brings them together, and the T cell does the rest to eliminate the target! This is one way we can use the immune system without a cellular therapy. No cells harvested from the patient. No personalized manufacturing. No lymphodepletion. Dosed like a drug. Most engagers today work through CD3. IN8bio is building one that works through gamma-delta T cells instead. More on why in the weeks ahead. #TCellEngager #Immunology #Biotech $INAB
