Well said on the role of somatic mutations in myeloid cells in Alzheimer's disease! (cell.com/trends/molecular-me…)
The CSF2RB A455D variant from our study is also a promising example. More studies using chimeric brain models with human microglia carrying somatic mutations are needed.
An idea we started in 2019 is now published in @NatureNeuro! Excited to share our new study: rdcu.be/eRvdA
Congrats to Mengmeng Jin @jin_mengmeng and our team! Heartfelt thanks to all collaborators for their support, and to the reviewers for their insightful comments!
Happy to share our Spotlight in @CellStemCell on advances in microglia replacement therapy (Wu et al., Science; Mader et al., Nature) and our perspectives on resilience variants & microglia reprogramming to enhance the “engineering brain sentinels” approach for brain repair.
This review was actually written once before (the section on methodological considerations for creating chimeric models is shown👇), but it didn’t make it to publication at the time.
I was inspired by Dr. Suchun Zhang’s work on differentiating human PSCs + transplanting them into mice, and Dr. Steven Goldman’s work on human oligodendroglia taking over mouse brains.
We discussed the idea of chimeric models in our 2014 Nat Commun paper 👇
Thrilled to share our review just published in @NeuroCellPress!
Big shoutout to Ava, Mengmeng , and our entire team!

Hacking the immune system could slow ageing — here’s how nature.com/articles/d41586-0…
But how to replace or rejuvenate the brain's immune cells, microglia? Hope our recent work can provide some clues.
