What happens when you actually DEGRADE KRAS G12D in pancreatic cancer?
At #AACRPanc26 we shared translational analyses of setidegrasib, the first-generation KRAS G12D degrader. @AstellasUS
15 paired tumor biopsies let us watch the biology change in patients:
Successful deep KRAS protein degradation.✏️
→↓ MAPK signaling
→ tumor-immune remodeling including Fas/FasL, Granzyme B+ CD8 T cells & PD-L1
→ distinct primary + acquired resistance biology!
→ rational mFOLFIRINOX and maybe immune combinations?
Parallel virtual patient and patient-derived organoids experiment from @orakldotbio predicting the human clinical trial outcomes independently.
What should we do next?
Patient → biopsies 💉→ biology 🧫→ model → combination → back to patient
