6) These SCFAs, w/ butyrate appearing to have the greatest potential role in immunity due to its palliative effect in inflammatory bowel diseases. In part this appears due to the improved quality of the epithelial barrier & diminished bacterial translocation and immune activation

Two routes to breaking the therapeutic ceiling in IBD, on one timeline. New drugs, if the trials succeed: the first anti-TL1As and obefazimod could reach Europe in 2028, and the first oral IL-23R peptide in 2029. Combinations: all three phase 3 readouts land in 2027 and 2028. The first two-in-one injection could follow around 2031. Next on Atomic IBD: the tablets, and whether they’ll arrive as partners rather than on their own.

Researchers at @WeillCornell @WCM_IBD found that patients with mild-to-moderate inflammatory bowel disease had a healthier gut microbiome after taking fluconazole, an antifungal medication. Treatment caused positive changes in both bacterial and fungal populations and was also associated with improved IBD symptoms. The findings now need to be confirmed in a larger, placebo-controlled trial but they offer a potential framework for developing more precise, mycobiome-guided treatments for IBD in the future. Free Access: Exploring the Link Between Environmental Exposures and Disease via Direct Challenge. Anna M. Erickson, Ian A. Myles, Jodi L. Blake, Cezmi A. Akdis, Hirsh D. Komarow Read the article here: doi.org/10.1111/all.70537 Mounting scientific evidence points to a link between environmental exposures and allergic diseases, as explained by the #epithelialbarriertheory. The clinical study THREAT, an acronym for Testing Harms Related to Exposure to Allergenic Toxins, compares the health impact of typical real-world indoor exposures to that of a controlled, healthier setting, such as free of toxic substances, reduced consumption of processed foods and purified air. Patient data will be collected on skin impedance, microbiome, cytokine profiles, and pulmonary function, among others. #Allergy_journal Read other articles published in #Allergy on the #epithelialbarrier here: journalallergy.com/barrier-a…
