Of course a first-gen therapy isn't going to be 100% flawless right out of the gate. But step back and look at what $MRNA $MRK just pulled off after a decade of grinding, a true paradigm shift in how we treat cancer. 1/2

For decades, every other personalized cancer vaccine failed in Phase 3. Delivering a 42% RFS improvement on top of Keytruda is a historic milestone that no one else has ever achieved. Baffling how people ignore this just to push short-sighted narratives. 2/2

Blown away by the lazy misinformation on $MRNA right now. People are screaming about deaths and missing OS data while ignoring what’s written right on the slide. You can't blame a vaccine for side effects that happened while patients were only on Keytruda pre-vaccine.

1/5 $MRNA $MRK I think some investors are overlooking an important point about INTerpath-001. This is an adjuvant melanoma trial, not a trial in patients with end-stage cancer. Patients had their tumors surgically removed, and the control arm received Keytruda.

5/5 There's a big difference between saying "OS benefit hasn't been established yet" and claiming "the vaccine doesn't improve survival." The first is scientifically accurate. The second is premature. Longer follow-up will tell us more. That's how adjuvant oncology trials work.

4/5 OS remains an important unanswered question. The current OS HR of 0.77 favors the combination, but the confidence interval is wide and the result is not statistically significant. That means the survival benefit is unproven, not that the vaccine has failed.

3/5 Meanwhile, INTerpath-001 met its primary endpoint for recurrence-free survival (RFS HR 0.58) and its key secondary endpoint for distant metastasis-free survival (DMFS HR 0.62). These are clinically meaningful results, not just statistical numbers.

2/5 Keytruda is already an effective immunotherapy. Patients in both arms can live for years, even after recurrence. With a median follow-up of just 24.6 months, immature overall survival (OS) data shouldn't be surprising. Demonstrating an OS benefit takes time.

$MRNA ESMO data, I think this is a much bigger deal than the market is giving it credit for. An RFS HR of 0.58 in Phase 3 is impressive, especially with a relatively modest increase in severe toxicity, seeing the Phase 2b results hold up in a larger trial is a big deal. 1/3

Sure, manufacturing costs, competition, and commercialization are real challenges. Nobody is denying that. But we're watching personalized mRNA cancer vaccines move from a promising concept to actual clinical evidence.2/3

That matters. And I think people are underestimating what this could mean beyond melanoma. Still early, but I'm genuinely impressed. 3/3

What makes $MRNA’s latest success so significant isn’t just the specific cancer it treated. It’s the paradigm shift it represents.1/3

Penicillin didn’t work against every bacterium, but it opened the door to countless antibiotics that later defeated many others. Moderna may have just opened a similar door for cancer treatment.2/3

The real value isn’t only what this means today, but what it makes possible tomorrow. That’s what makes this achievement so remarkable.3/3

$MRNA deal has some drawbacks, but the benefits clearly outweigh them. Moderna locked in $2.6B at 0% through 2032 just before global bond yields surged. With the U.S. 10-year near 4.8%, the timing looks even smarter now.
