📢 I am so incredibly happy and proud about this one! 🔟+ years in the making, 2,5 years since first submission, 1⃣ year since we posted it @biorxivpreprint, it finally found it’s home @NatureCellBio! Open access 👇 nature.com/articles/s41556-0… Detailed 🧵down below ⏬
⚡️New @biorxivpreprint from our group⚡️ In this manuscript, we provide answers to the long-standing question "Where is mTOR and what is it doing there?", further complementing it with insights about "WHY is it there in the first place?" biorxiv.org/content/10.1101/… Thread below👇

GRASP55-deficient cells demonstrate lysosomal enzyme missorting (ending up into the extracellular space), #lysosome dysfunction and bloating, and perturbed lysosomal #mTORC1-TFEB/TFE3 signaling (while leaving non-lysosomal mTORC1 signaling intact).

Mechanistically, GRASP55 is required for the Golgi localization of the complete GOLPH3-LYSET-GNPTAB complex. In the absence of GRASP55, mislocalization of #LYSET and #GNPTAB is linked to their destabilization, in turn causing defective tagging and missorting of lysosomal enzymes.
