Our latest preprint "Sequential structure probing of cotranscriptional RNA folding intermediates" is on bioRxiv. biorxiv.org/content/10.1101/… Led by @CourtneySzyjka and anchored by Skyler Kelly, this preprint describes a method for detecting cotranscriptional RNA folding transitions.

And in the presence of ZMP, we also observed both folding transitions and (also as expected) clear signatures of ZMP binding.

We applied this method to 5 known folding transitions, which allowed us to assess it's generalizability and target RNA-specific challenges that may arise, which are described in detail in the preprint.

Fresh preprint! Cool RecBCD work led by Alessia Lepore in @MEKlab. @ojp25 contributed from my lab. Nice imaging + mechanistic (SOS!) insight. @BBSRC-funded. In vivo single-molecule imaging of RecB reveals efficient repair of DNA damage in Escherichia coli biorxiv.org/content/10.1101/…

We have received a lot of requests for help with making streptavidin affinity grids. We are happy to finally share our protocol for robust in-house fabrication of streptavidin affinity grids. Stay tuned for the accompanying video that is coming soon! app.jove.com/t/66197/strepta…

Want to design genetically encoded inhibitors of your favorite protein? Thrilled to announce our latest work w/ @SassSeabass, @keating_lab, & @GeneWeiLiLab, where we leverage massively parallel AlphaFold predictions to discover peptide inhibitors. biorxiv.org/content/10.1101/… (1/n)

We applied this to a ZTP riboswitch variant library to investigate the mechanisms by which variants that match the consensus sequence and structure misfold. Panel c illustrates the ZMP-mediated antitermination response for the 32,768 variants in the library!

We designed the TECdisplay platform to be broadly accessible and easy to use, so our hope is that this approach democratizes the development and application of high-throughput RNA biochemical assays.
