Most friends know my story, but new followers probably don't - short version:
-Am a Wall Street Nobody, started investing in 2008 with no one to teach me & made every mistake in the book (& then some, very lucky to have survived them)
-Writing on SA been a hobby for long time (comments were brutal, forced me to keep improving & showed just how little I truly know about this sector)
-After university went to Peru (figured have some fun before getting a "real job", 6 months turned into 15 years there 😂
-Committed career suicide by quitting a lucrative finance job to instead teach English at university. One night out at a club with friends, met a salsa dancer then turned into a 10yr relationship/7yr marriage
-Along the way, used clinical-stage biotech wins to start a couple successful businesses (restaurant & fashion), also bought a condo to diversify. Biggest binary loss was ironically not biotech but instead a divorce Xmas 2021 (such is life sometimes, thankful for what I got)
-Started from square one again as a biotech hobo, spending about 3 months in each country for 2+ years now. Life often turns out differently than we expect, but it's still pretty grand and good reminder that God always has a plan for each of us 🤙
Normally don't deviate into the personal, but hope it gives perspective for anything I post in the future
Cheers my friends 🙂 – at Limón, Costa Rica
Really great call - my favorite theme was how to benefit from our rough patches & countering a negative mindset 👍
Thanks to all who kept the conversation flowing, including @WassimLaroussi3 @lfholt @Sanctuary_Bio @JoseRestonVA and others not on X
Have a great weekend all 🤝

"totality of the strategy" was @lfholt's phrase, I liked it
Zero Ego trades= my favorite stock today can be the one I sell tomorrow
If conviction level changes, thesis takes a hit or via our Portfolio Cagematch exercise another name looks more attractive
All about the "totality of the strategy", making the best decisions I can each day
Fwiw, $IOVA back on my DD list (expect the weakness to continue for some time though)
Perhaps one man's trash will be another's treasure at the right price
TILVANCE-301 frontline melanoma trial has March 2028 primary completion date (ORR interim prior)
IL-12 tethered TIL unlocks cold tumors, next-gen IL-2 reduces AE burden from conditioning
Expansion indications of endometrial cancer & sarcoma should've made good progress by then
110 active ATC sites by year-end (infrastructure = significant strategic value to larger players vs having to scale up from scratch, can always improve the tech along the way)
Ideal scenario= pendulum swings to being out of favor again, overreaction lasts well into 2027 before reverting
Whether this name or others on radar,
Goal= let optimal setups come to me (not chasing)
$IBB quietly breaking out 🧐

I get many wrong my friend 🙂
$AFMD was certainly painful
I've been blocked by a lot of people for my devil's advocate posts, is what it is.
Occasionally just a misunderstanding - will continue to focus on adding value where we can, the rest takes care of itself.
$IOVA - greatly respect longs like @kainvests that instead of stubbornly clinging to their original thesis, are quick to admit where it's been negatively impacted
1) "Durability has substantially regressed" - clear that low dose lymphodepletion adversely affecting DoR
2) Mgmt misled investors going into the update, plain & simple
(coming from someone whose weakness is also being slow to admit where he's wrong... and pays dearly for it - I could learn a thing or two!)

Attaching as a visual reference.
Yes, we have several ongoing responders after three or four years post-infusion, but way too many ended much earlier, with progression or study discontinuation around 6-18 months after infusion...
In short, TIL is not delivering stable/uniformly prolonged responses across the responding population.
Durability seems to be around 12 months, at best. But we don't have confirmation about mDOR, which is concerning in itself. They were able to provide for an n of 39 and all of a sudden they can't for an n of 69 patients? Right!
Unless I've misunderstood something, the durability has regressed substantially and approval based on positive subgroups will need way more time than expected. We are definitely not getting approval in 2027.
2 eye prosthesis taken care of (retinoblastoma), MRIs next 🤝

One last point- if the data were strong & this were truly a "sell the news" reaction, I'd be in or at least seriously considering (Playbook)
However, there's very low probability of accelerated approval as it stands (in addition to concerns raised by relying on subset analysis)
Scenario where they get a lung approval which leads to accelerating revenue growth that paves the way for sarcoma & endometrial cancer = highly attractive to my eyes
OTOH, one where lung's in limbo and we have to wait multiple years for approvable data sets in other indications = type of situations I try to avoid
My two cents... GLTA (& be careful, a lot of unfounded cheerleading out there today)

Iovance's updated IOV-LUN-202 data (n=69 non-squamous mNSCLC post-chemo/IO) showed 24.6% ORR (15 confirmed = 21.7% cORR). That roughly matches REVEL's 22.9% for doce+ramu (pre-IO era). Post-IO real-world doce/ramu ORRs often run higher (28-40% range).
Prior smaller set had mDOR not reached at 25 mo follow-up; CEO noted current swimmer plot implies ~1 yr and data still immature. No PFS released. Safety called consistent with reduced-dose lymphodepletion (prior hold was 2023, lifted after protocol changes). Company is pivoting to a KRAS-WT/HER2-WT/smoker subgroup (59% ORR) for the new POSITILITY cohort.

but that all said? Yes I stand by that. Because that's the goal right? FI and what not...
I'm not saying it's easy. It shouldn't be. But I think esp as you get experienced, this is a question you have to think more about.
