First announced patient dosing of an in vivo prime editing treatment (to correct ATP7B H1069Q, one of the most common genetic causes of Wilson disease). There are now at least three prime editing clinical trials underway—for CGD (ex vivo, with positive clinical outcomes), for Wilson disease (in vivo), and for alpha-1 antitrypsin deficiency (AATD, in vivo).

Awesome PI and a fantastic lab to apply for technology development!
Replying to @AllenYJiang
To unlock the full clinical potential of highly stable editors like PE8, we need advanced delivery tech. I'm excited to tackle this by engineering non-viral delivery systems in my independent lab at @UclaCBE starting July 2027! Recruiting for Fall '27: sites.google.com/g.ucla.edu/…

It is incredibly exciting to see this story finally out nature.com/articles/s41551-0…. I feel very fortunate to have completed this work with my post-doctoral mentor @davidrliu's support. This study would not have been possible without our outstanding collaborators Cat, Joe, Nancy, Guangping, and Max. I also want to give a special shout-out to my undergraduate student @jordynduby, who contributed to this project and is now an MD/PhD student at @PennMedicine. This is only the beginning of precision gene editing for peroxisome biogenesis disorders and other genetic diseases. As highlighted in the @broadinstitute article broadinstitute.org/news/base…, insights from this study helped inform the use of the TadA-8e-V106W deaminase in the base editor used to treat baby K.J. Muldoon’s CPS1 deficiency, even before this paper was published. Now at @PittTweet @Pitt_OPH, my lab is building novel genome engineering and delivery technologies to enable more efficient, cost-effective, and safer medicines to advance human health. Stay tuned.
Today in @natBME we report an in vivo base editing strategy that corrects a common disease-causing mutation and rescues pathology in a mouse model of Zellweger spectrum disorder (ZSD) and restore peroxisomal function in patient derived cells. This work highlights the promise of precision genome editing for treating peroxisomal biogenesis disorders. 1/13

One of the most accurate descriptions of the unbearable situation - in which many Gene Therapies and CRISPR Gene Editing therapies fail to reach patients only due to economical and regulatory hurdles 🧵👇, can be found in this statement - which was made during the recent FDA's Cell and Gene Therapy roundtable, by Prof. @davidrliu the inventor of Base and Prime Editing and the Co-founder of $BEAM & $PRME. During his excellent presentation Prof. Liu emphasised the many challenges facing the field including the lack of commercial potential in many cases due to the small number of patients and the need to approve each treatment for one of the 10,000 different rare genetic diseases separately. According to Prof Liu if the sufficient support will be provided by the government - both regulatory and financially, the BioTech and Pharma ecosystem would be able to generate personalised and customised genetic treatments for 1,000 patients each year by 2030, but most importantly would be able to unlock and pave the way for many more patients and treatments down the road. It’s time to accelerate and bring hope to millions of patients and their families worldwide!
1/In one of the best articles that I’ve read recently - Washington Post’s excellent science reporter @Carolynyjohnson sheds light on a less known aspect of the Gene Therapy and CRISPR Gene Editing field. Although many Gene Therapies and CRISPR Gene Editing therapies are clinically proven and could generate major health benefits for patients, they are still being discarded and fail to reach patients only due to economical and regulatory hurdles. In her excellent piece Carolyn Johnson brings not only the voice of the physicians and of the patient community but also that of the scientists who are developing the genetic treatments for these rare genetic diseases including @kiranmusunuru who treated the baby KJ Muldoon with a custom made Base Editing treatment and that of Professor @davidrliu - the inventor of Base and Prime Editing and the co-founder of Beam Therapeutics (NASDAQ: $BEAM) and Prime Medicine (NASDAQ: $PRME) - all agreeing that the current situation is both unacceptable and heartbreaking! An outstanding example for this anomaly - that existing and clinically proven medical treatments for rare genetic diseases are not being offered to patients who are desperate for a cure, can be seen in many cases such as $PRME recent decision to discontinue its clinically proven PM359 program for treating CGD - a rare inherited immune deficiency disorder. Custom Gene Therapy and Gene Editing treatments are not a vague or futuristic concept but a reality! The regulators must get involved and intervene in order to immediately solve this issue which affects more than 300 million patients with rare genetic diseases and their families worldwide!! 🧵👇 $XBI

I'm thrilled to share that I’ve officially launched my lab this week at the University of Pittsburgh Department of Ophthalmology/UPMC Vision Institute @Pitt_OPH ! I’m incredibly grateful for the mentorship and support I’ve received throughout my journey from @davidrliu , @ESontheimerLab , and Chonghui Cheng during my postdoctoral and graduate training. I’ve also been very fortunate to collaborate with outstanding scientists like Joe Hacia, Cat Lutz, Nancy Braverman, and Job Dekker, whose partnership has been truly invaluable. A heartfelt thank you to all my past colleagues and those I’ve met during the faculty interview process. The mission of the Gao Lab is to uncover fundamental biological mechanisms and translate these discoveries into transformative therapies that advance human health. We are hiring at all levels! If you're passionate about driving the future of genomic medicine, I’d love to hear from you. Learn more about my work: lnkd.in/eZpAgYD9 and please reach out to me via [email protected] More updates coming soon—stay tuned!

Another medical milestone! The first patient treated with a prime edited therapeutic shows positive clinical outcomes consistent with effective rescue of his chronic granulomatous disease (CGD), a debilitating and life-shortening primary immunodeficiency. At least 19 base editing and prime editing clinical trials have begun, with at least 7 trials reporting clinical outcomes (highlighted in green in the table below). They treat a wide range of diseases both in vivo and ex vivo. And while the U.S. support for science is now in crisis, that there are more base editing and prime editing clinical trials serving patients outside of the U.S. (13) than within the U.S. (9), despite the fact that these technologies were developed in our lab in the U.S. investors.primemedicine.com/…

In a medical milestone, a customized base editor was developed, characterized in human and mouse cells, tested in mice, studied for safety in non-human primates, cleared by @US_FDA for clinical trial use, manufactured as a complex with an LNP, and dosed into a baby with a severe, rapidly progressing genetic disease... all in an astounding 7 months. Best of all, the infant patient shows apparent benefit. Congratulations to @kiranmusunuru, Rebecca Ahrens-Nicklas, and other team members for this heroic and inspiring effort, which has implications for the hundreds of millions of patients that suffer from thousands of genetic diseases. drive.google.com/file/d/1Jfk…
