10/ APEX linked luminal programs to favorable outcomes and EMT signatures to poor outcomes. In enfortumab vedotin–treated bladder cancer, 58% of patients with high APEX-predicted NECTIN4 responded, compared to 0% with low NECTIN4.
Even after adjusting for tumor fraction, copy number, and clinical covariates, APEX-derived NECTIN4 remained independently associated with improved survival. Our preliminary data suggests that plasma-inferred NECTIN4 may outperform tissue NECTIN4 IHC in this task.
Thanks to @UCSFCancer for covering our study which showed that a PPARg agonist rosiglitazone (an old diabetes drug) can modulate NECTIN4 & enhance activity of a new CAR T in #bladdercancer @CChuMD @koshkin85 @DrTFriedlander @SimaPorten @UCSFUrology ucsf.edu/news/2025/09/430701…

We are happy to share our new study published today @JCO_ASCO (👉 ascopubs.org/doi/10.1200/JCO…) on the role of NECTIN4 amplifications to predict responses to Enfortumab Vedotin treatment in metastatic urothelial cancer.
With a huge contribution of our multinational team we uncovered the role of NECTIN4 amplifications in enhancing the efficacy of Enfortumab Vedotin (EV), a cutting-edge treatment for metastatic urothelial cancer (mUC).
With NECTIN4 amplifications positioned as a potential genomic marker to predict responses to EV in mUC patients, let's explore how these findings could translated to other cancer entities as well!

Great overview on UC pathology @Markuseckstein3 🔥 important to keep these pathways of UC carcinogenesis in mind for ADC targets as well, as NECTIN4 is overexpressed in luminal tumors as nicely demonstrated by @jonchou05 pubmed.ncbi.nlm.nih.gov/3410… in @CCR_AACR @urotoday @OncoAlert

Our latest paper ist out in @Histo_Journal ! We comprehensively studied the membranous protein expression of the two important #ADC targets TROP2 and NECTIN4 (surfaceome 🏄♂️). it is the first paper on protein expression in a large cohort of over 200 muscle-invasive and metastic carcinomas of the urinary bladder. 😊
